Treatment was initiated in the schizont stage [42 hours post invasion (hpi)] and cultures were sampled based on morphology of the untreated parasites at t1 = 19hpi, t2 = 27hpi and t3 = 34hpi of the next cycle approximately. RNA was extracted and a common reference RNA pool was prepared. cDNA was prepared as described (Bozdech, 2003). Sample cDNA was labeled with Cy5 and the common reference with Cy3. Oligonucleotide hybridization was perfomed at 65 C overnight, followed by washing and scanning with an Axon GenePix 4000A scanner.
